F4V format does not contain ActionScript 3 accessible cue points.

You have to add them (in AS3 or in the Component Inspector panel), use some net connection method, or else use FLV format for the video. This is true even though Media Encoder allows you to add/change cue points when F4V output is selected. This only took a day to find an answer. Mostly because when you do a search for “actionscript 3 FLVPlayback cue points”, you get all the tutorials and docs talking about how to set up for FLV cue points.

Other options are using F4V and NetConnection, using F4V and netStreamClientClass, or following Adobe’s guidelines concerning F4V cue points.
